The truth of the day
This verse comes at the close of the conquest narrative, after the land has been distributed to the tribes. It is a summary statement, and its claim is total: nothing failed of any good thing God had spoken.
This is a promise already kept in Israel's own history, offered as evidence for trusting the promises still ahead. If God's word proved reliable about the land, it can be trusted about the greater promises still unfulfilled at that point in the story.

Deeper insight
What the passage says in its own setting
Joshua 21:45 stands at a structural hinge in the book. Chapters 13 through 21 have been an extended, almost tedious list of tribal land allotments, easy to skim past. This verse is the narrator stepping back to say what all those lists mean: every promise made to the patriarchs about this land has now come to pass.
The claim is repeated with slight variation in Joshua 23:14, near the end of the book, where Joshua tells the elders not one word has failed of all the good things Yahweh promised, and warns that the same God who keeps good promises will also keep his warnings if they turn away. Fulfilled promise is presented as grounds for continued obedience, not a reason to relax.
